How It Works:

The assay relies on the diazotisation of bilirubin in the presence of diazotised sulphanilic acid. Caffeine acts as an accelerator in total bilirubin estimation, while direct bilirubin reacts directly. The intensity of the pink-coloured complex formed is proportional to the bilirubin concentration and is measured photometrically.
