Intended Use

The Amylase Reagent Kit is intended for the quantitative determination of α-amylase in serum and urine, supporting the clinical assessment of pancreatic function and other digestive conditions.

Assay Principle

The method is based on enzymatic cleavage of CNPG3 (2-chloro-4-nitrophenyl-α-maltotrioside) by α-amylase, producing a chromogenic complex. The intensity of the resulting colour is directly proportional to the amylase activity and is measured spectrophotometrically at 405 nm.

Kit Components

  • R1 (Buffer Reagent): 4 × 25 mL

  • R2 (Substrate Reagent – CNPG3): 1 × 25 mL

Storage & Stability

  • Storage Condition: 2°C to 8°C
  • Stability: Stable until the expiry date if unopened and stored as recommended.
  • Onboard Stability: Reagents remain stable for 7 days on the analyser at 2°C to 10°C.

Sample Requirements

  • Specimen Types: Serum and urine
  • Handling: Avoid haemolysed samples. Samples should be tested promptly or stored at 2–8°C for up to 7 days or at -20°C for longer storage.

Performance Characteristics

  • Linearity: Up to 2000 U/L
  • Sensitivity: Detects amylase activity ≥ 1 U/L
  • Precision:
  • Intra-assay CV: < 2%

  • Inter-assay CV: < 3%

  • Interference: Haemoglobin up to 500 mg/dL and bilirubin up to 20 mg/dL do not interfere significantly.

Quality Control

Use normal and abnormal controls with each batch to ensure result reliability. Compatible with most open-system analysers.

FAQs: Amylase Reagents

What is the CNPG3 method used in this kit?

The CNPG3 method is a chromogenic substrate-based enzymatic reaction that provides a rapid and specific colourimetric reading of α-amylase activity, measured at 405 nm.

What types of samples can be tested with this kit?

This assay is suitable for human serum and urine samples. For accurate results, avoid haemolysed samples and follow proper storage guidelines.

What are normal amylase levels?

According to the NHS, normal amylase levels in blood range from 23 to 85 U/L, though reference ranges can vary by lab.

Can these reagents be used on all clinical chemistry analysers?

Yes, the kit is compatible with most open-system analysers used in diagnostic laboratories.

How should I store the kit once opened?

Store the reagents between 2°C and 8°C. Onboard stability on automated analysers is up to 7 days.
