HDL Cholesterol Reagent Kit – Direct Precipitation Method for Serum Lipid Profiling
Vanguard’s HDL Cholesterol Reagent Kit is designed for the quantitative estimation of high-density lipoprotein cholesterol (HDL-C) in human serum. Using the phosphotungstate-magnesium chloride precipitation method, the kit enables selective separation of HDL from other lipoproteins for accurate lipid profiling.
This liquid-stable reagent is easy to use, suitable for manual or semi-automated analysers, and ensures reliable results with minimal preparation. Ideal for clinical laboratories conducting routine lipid panel assessments.

Key Features
- Method: Precipitation with Phosphotungstic acid/MgCl₂
- Sample Type: Human serum
- Measuring Wavelength: 546 nm (acceptable range: 500–600 nm)
- Reagent Stability: Liquid stable
- Linearity: Up to 120 mg/dL
- Storage: 2–8°C, protected from light
Kit Components
- R1 (Precipitating Reagent): 1 x 50 mL / 2 x 50 mL
- R2 (Cholesterol Reagent): 1 x 100 mL / 2 x 100 mL
Intended Use
The HDL reagent kit is intended for the quantitative determination of HDL cholesterol in human serum samples. It forms part of a broader lipid profile and is used in the diagnosis and monitoring of cardiovascular conditions.

Test Principle
Non-HDL lipoproteins are precipitated by phosphotungstic acid and magnesium ions, leaving HDL in the supernatant. The HDL cholesterol concentration is then measured enzymatically using a CHOD-PAP method.
Storage and Stability
- Reagents are stable for 24 months at 2–8°C.
- Do not freeze.
- Ensure reagents are kept away from direct light and contamination.
Test Procedure Summary
- Mix 0.2 mL of serum with 0.5 mL of precipitating reagent (R1).
- Allow to stand for 10 minutes at room temperature.
- Centrifuge at 4,000 rpm for 10 minutes.
- Collect supernatant and measure HDL cholesterol using reagent R2 at 546 nm.
FAQs
What is HDL cholesterol and why is it important?
HDL (high-density lipoprotein) is often referred to as “good cholesterol” because it helps remove excess cholesterol from the bloodstream.
What method does this kit use to measure HDL?
It uses a precipitation method with phosphotungstic acid and magnesium, followed by enzymatic colorimetric measurement (CHOD-PAP).
What type of analyser can I use with this kit?
The kit is compatible with manual and semi-automated analysers.
Is it suitable for routine use in clinical labs?
Yes, the kit is designed for routine lipid panel testing and delivers consistent and reliable results.
What is the linearity range for this test?
The assay is linear up to 120 mg/dL.
What kind of samples can be used?
This test is validated for use with human serum samples only.
Is this product CE certified?
Yes, the kit meets CE standards for in vitro diagnostic use.
